韓·日 공동연구팀, 세포 소기관별 온도 빛으로 볼 수 있는 방법 개발

【브레이크뉴스 포항】박영재 기자=POSTECH(포항공과대학교, 총장 김무환)은 화학과 장영태 교수(기초과학연구원(IBS) 복잡계 자기조립 연구단 부단장)·IBS 샤오 리우(Xiao Liu) 박사 연구팀이 일본 카나자와대(Kanazawa) 사토시 아라이(Satoshi Arai) 교수·타케루 야마자키(Takeru Yamazaki) 학생과의 공동연구를 통해 세포 소기관별 형광 온도계를 개발했다고 1일 밝혔다.

이 연구성과는 국제 학술지 ‘머터리얼즈 투데이 바이오(Materials Today Bio)’에 최근 게재됐다.

 

세포 반응과 밀접하게 연관된 세포기관의 온도 변화는 복잡한 세포를 이해하는 중요한 실마리가 될 수 있다. 다만, 각기 다른 세포 소기관의 미세한 온도 변화를 알기 위해선 세포 소기관별 온도계의 개발이 필수적이다. 앞서 형광 온도계가 개발됐지만, 한두 가지의 세포기관에만 적용할 수 있다는 한계가 있었다.

 

연구팀은 형광 온도계로 세포막, 핵, 골지체, 소포체, 미토콘드리아, 지방방울, 리소좀을 망라하는 거의 모든 세포기관의 온도 변화를 시각화하는 데 성공했다. 한겨울에도 체온을 유지하는 데 중요한 갈색 지방세포 속 미토콘드리아나 소포체 등 다양한 세포 소기관의 온도를 정량적으로 영상화할 수도 있었다.

 

이 연구성과는 대부분의 세포 소기관에 적용할 수 있는 최초의 소형 분자 형광 온도계 팔레트로 주목을 받는다. 이를 토대로 향후 새로운 형광 온도계를 개발할 수 있을 뿐만 아니라, 열생물학에 대한 이해를 한층 높일 수 있을 것으로 기대된다.

 

한편, 이 연구는 IBS, 일본과학기술진흥기구, 일본학술진흥회의 지원을 받아 이뤄졌다. 연구팀은 이번 연구성과를 POSTECH·IBS·가나대와대 대학 간 특허로 출원했다.

POSTECH (Pohang University of Science and Technology, President Kim Moo-Hwan) is a research team led by Professor Young-Tae Jang (Assembly Director of Complex Systems Self-Assembly Research Center, Institute of Basic Science (IBS)) and Dr. Xiao Liu's research team from the Department of Chemistry, Professor Satoshi Arai of Kanazawa University, Japan ·The company announced on the 1st that it had developed a fluorescence thermometer for each organelle through joint research with student Takeru Yamazaki.

 

  This research result was recently published in the international academic journal ‘Materials Today Bio’.

 

  Temperature changes in organelles, which are closely related to cellular responses, can be important clues in understanding complex cells. However, it is essential to develop a thermometer for each organelle in order to know the minute temperature changes of different organelles. A fluorescent thermometer was previously developed, but there was a limitation in that it could be applied only to one or two organelles.

 

  The research team succeeded in visualizing temperature changes in almost all organelles including the cell membrane, nucleus, Golgi apparatus, endoplasmic reticulum, mitochondria, fat droplets, and lysosomes with a fluorescent thermometer. It was also possible to quantitatively image the temperature of various organelles, such as mitochondria and endoplasmic reticulum in brown adipocytes, which are important for maintaining body temperature even in midwinter.

 

  This research achievement draws attention as the first palette of small molecule fluorescence thermometers applicable to most organelles. Based on this, it is expected that it will be possible to develop a new fluorescence thermometer in the future, as well as to further enhance the understanding of thermobiology.

 

  Meanwhile, this study was conducted with the support of IBS, the Japan Science and Technology Promotion Agency, and the Japan Association for the Promotion of Science. The research team applied for a patent between POSTECH, IBS, and the University of Ghana.
