홍합접착단백질과 히알루론산으로 관절염 치료 줄기세포 접착제 개발

보건복지부 보건의료기술개발사업(세포 기반 첨단 조직공학 융복합 기술 실용화 지원)의 지원으로 진행된 이 연구는 화학공학 분야에서 영향력이 높은 학술지인 ‘케미컬 엔지니어링저널 (Chemical Engineering Journal)’에 게재됐다.

 

연골은 외부 충격으로부터 뼈를 보호하고, 관절이 원활하게 움직이도록 돕는다. 그런데 연골은 자연적인 치유 능력이 없어 염증이 생기거나 연골이 손상되는 경우, 줄기세포 이식을 통해 연골 재생을 유도한다. 하지만 연골의 표면이 매우 매끄럽고, 연골 주위에 끈끈한 윤활액이 있어 이식된 대부분의 줄기세포는 이식 초기에 산산이 흩어져 사라져 치료 효과가 미미하다.

 

연구팀은 이번 연구에서 홍합에서 유래한 접착단백질과 고분자량의 히알루론산을 결합한 액상형 접착제를 개발했다. 홍합 단백질과 히알루론산은 서로 반대의 전하를 띠고 있어 둘 사이에는 서로를 끌어당기는 힘(정전기적 인력)이 발생하게 된다. 연구팀은 이 강력한 힘을 이용하여 끈적끈적한 점성은 있지만 물에서도 와해되지 않는 고점도의 액체를 만들었다. 줄기세포를 이 액체에 넣어 원하는 부위에 단단하게 고정할 수 있는 줄기세포 이식용 접착제를 만든 것이다.

  

이어, 연구팀은 토끼의 손상된 연골 부위에 액상형 접착제에 들어있는 줄기세포가 풀어지지 않은 상태로 고르게 이식됨을 확인했다. 또, 줄기세포가 오랫동안 안정적으로 고정되어 있어 손상된 연골이 정상 연골로 재생되는 등의 치료 효과도 확인했다.그리고, 연구팀이 개발한 액상형 접착제는 물리 · 화학적으로 추가적인 공정이 필요하지 않은 자연 접착제라는 것도 큰 장점이다.

 

이번 연구를 이끈 차형준 교수는 “대한민국 원천소재인 홍합접착단백질을 활용하여 줄기세포 치료 효과를 크게 증대시킬 수 있었다”며, “주사가 가능한 제형이기 때문에 내시경과 유사한 관절경을 통한 줄기세포 이식에 활용한다면 보다 효율적으로 연골을 치료할 수 있을 것”이라는 기대를 전했다.

 

한편, 홍합접착단백질 소재 기술은 ㈜네이처글루텍에기술이전이 완료됐고, 본 공동연구를 통해 개발된 관절염 치료 줄기세포 접착제는 ‘카티픽스(CartiFix)’라는 제품명으로 조만간 임상을 진행할 예정이다.

 Development of stem cell adhesive for arthritis treatment using mussel adhesive protein and hyaluronic acid

This research, which was supported by the Ministry of Health and Welfare's health and medical technology development project (support for commercialization of cell-based advanced tissue engineering convergence technology), was published in the Chemical Engineering Journal, an academic journal with high influence in the field of chemical engineering.

 

  Cartilage protects bones from external impact and helps joints move smoothly. However, cartilage has no natural healing ability, so when inflammation occurs or cartilage is damaged, cartilage regeneration is induced through stem cell transplantation. However, since the surface of the cartilage is very smooth and there is a sticky lubricating fluid around the cartilage, most of the transplanted stem cells are scattered and disappear in the early stages of transplantation, and the treatment effect is insignificant.

 

  In this study, the research team developed a liquid adhesive that combines mussel-derived adhesive protein and high molecular weight hyaluronic acid. Mussel protein and hyaluronic acid have opposite charges, so a force (electrostatic attraction) that attracts each other occurs between the two. The research team used this powerful force to create a high-viscosity liquid that is sticky but does not disintegrate even in water. Stem cells are put into this liquid to make an adhesive for stem cell transplantation that can be firmly fixed to a desired area.

 

  Subsequently, the research team confirmed that the stem cells contained in the liquid adhesive were evenly transplanted to the rabbit's damaged cartilage without loosening. In addition, the stem cells were stably fixed for a long time, and the treatment effect, such as regeneration of damaged cartilage into normal cartilage, was also confirmed. Another great advantage of the liquid adhesive developed by the research team is that it is a natural adhesive that does not require additional physical or chemical processes.

 

  Professor Cha Hyeong-joon, who led this study, said, “We were able to greatly increase the effect of stem cell treatment by using mussel adhesive protein, a material sourced from Korea. If we do, we will be able to treat cartilage more efficiently.”

 

  Meanwhile, technology for mussel adhesive protein material technology has been transferred to Nature Glutech Co., Ltd., and the arthritis treatment stem cell adhesive developed through this joint research is expected to undergo clinical trials soon under the product name 'CartiFix'.
