 POSTECH(포항공과대학교, 총장 김무환) 연구팀이 홍합이 만들어 내는 강력하면서도 인체에 무해한 생체접착소재인 홍합접착단백질을 이용해 혈관 형성 인자들을 시공간적으로 효율적으로 전달할 수 있는 접착패치 플랫폼을 최근 개발했다고 6일 밝혔다.

POSTECH 화학공학과 차형준 교수, 박태윤 박사 연구팀은 세포배양공정을 통해 대량생산이 가능한 홍합접착단백질을 기반으로 형성할 수 있는 두 가지 제형인 코아서베이트 기반 마이크로입자와 광가교 기반 하이드로젤을 이용하여 약물전달 접착패치를 개발했다.

 

그리고 효용성 검증을 위해 중증 모델로 랫트(Rat)를 이용한 심근경색 모델과 자가 치유 불가 피부 손상 모델에서 효율적인 신생 혈관 형성을 비롯하여 기능적인 조직재생 효능을 실험적으로 검증했다.

 

연구팀은 홍합접착단백질과 히알루론산의 정전기적 인력으로 형성되는 코아서베이트 액상체를 만드는 과정에서 혈관 형성 후기에 필요한 혈소판 유래 성장인자(Platelet Derived Growth Factor; PDGF)를 포집하고 순간적으로 가교1)하여 마이크로입자를 제작했다.

 

또한, PDGF가 포집된 마이크로입자와 혈관 형성 초기에 필요한 혈관 내피세포 성장인자(Vascular Endothelial Growth Factor; VEGF)를 함께 홍합접착단백질 기반 광가교 하이드로젤에 동시에 포집하고, 두 인자들을 쉽고 빠르게 공간적으로 분리하여 탑재했다.

 

이렇게 만들어진 효율적 신생 혈관 형성 유도 접착패치 플랫폼은 PDGF가 VEGF보다 작은 크기에도 불구하고 시간적으로 1.9배나 늦게 분비되어 전달되는 것으로 나타났다. 뿐만 아니라, 이 플랫폼은 광가교가 순간적으로 일어날 수 있도록 해 평평하지 않는 곡면에서도 쉽게 접착될 수 있음을 확인했다.

 

홍합접착단백질을 꾸준히 연구해 온 차형준 교수는 “대한민국 원천소재인 홍합접착단백질의 제형적 특징을 활용해 혈관 형성 인자들의 시공간적인 전달을 효율적으로 할 수 있는 새로운 플랫폼을 개발했다”며, “혈관 형성이 꼭 필요한 실제 심근경색 동물모델과 중증 피부 손상 모델에 적용하여 효과적인 신생 혈관 형성을 넘어서 기능적 회복까지 확인한 것에 커다란 의의가 있다”라고 연구의 의미를 밝혔다.

 

또한 “이와 관련된 비슷한 환경의 만성 및 허혈성 질환에도 성공적으로 적용할 수 있을 것”이라며 “이 플랫폼은 인체에 무해한 생체적합성 바이오소재를 이용한 것인 만큼 신생 혈관 형성 치료제 시장의 핵심적인 역할을 할 수 있을 것이다”라며 기대감을 밝혔다.

 

한편, 홍합접착단백질 기반 광가교 하이드로젤과 코아서베이트 액상체 제형기술은 ㈜네이처글루텍에 기술이전을 완료, 현재 상용화가 추진되고 있다.

 

이번 연구 결과는 바이오소재 분야 최고 권위지인 ‘바이오머터리얼즈 (Biomaterials)’에 게재됐으며, 보건복지부 및 한국보건산업진흥원에서 지원하는 보건의료기술개발사업과 과학기술정보통신부와 한국연구재단에서 지원하는나노·미래소재원천기술개발사업의 일환으로 수행됐다.
