Development of a stem cell-based supernatural bone spheroid differentiation promotion platform for ‘cartilage regeneration’

POSTECH (Pohang University of Science and Technology, President Kim Moo-Hwan) is a joint research team led by Professor Dong-Seong Kim of the Department of Mechanical Engineering at POSTECH, Professor Seong-Jin Lee of the integrated course, Professor Ju-Hyun Ju of Rheumatology at the Catholic University of Korea Seoul St. It announced on the 23rd that it has developed a cell culture platform based on nanofiber wells that promotes permeation and has significantly improved the differentiation efficiency of stem cells into hyaline cartilage spheroids.

The results of this research were recently published in Biofabrication, a leading international academic journal in the field of bioprocessing technology.

For effective cartilage regeneration, stem cell spheroids are the main component of healthy cartilage, not fibrous cartilage, and must be differentiated into supercartilage with excellent therapeutic effect. When the stem cell spheroids were differentiated into cartilage in a conventional impermeable cell culture vessel, the differentiation efficiency into supercartilage was inhibited due to the heterogeneous chondrogenic differentiation environment around the spheroids.

To overcome this drawback, the research team developed a micro-deep drawing process for the nanofiber membrane and fabricated a permeable nanofiber well that mimics the periosteum. Oxygen and growth factors essential for cartilage differentiation can smoothly permeate through the permeable wall of the nanofiber well, making it possible to supply a uniform chondrogenic differentiation environment to the stem cell spheroids, which promotes the differentiation of stem cell spheroids into supercartilage. succeeded in doing

In addition, the research team confirmed that the cartilage differentiation period, which took 20 days in the existing impermeable cell culture vessel, was shortened to 14 days using the nanofiber well, and the cartilage function was greatly improved. Finally, when supercartilage spheroids produced using nanofiber wells using a cartilage defect mouse model were transplanted into the cartilage damaged area, the transplanted area was regenerated into a tissue similar to supercartilage rather than fibrocartilage after 8 weeks. to confirm the therapeutic effect.

Professor Dongseong Kim of POSTECH said, “Inspired by the supply of oxygen and nutrients to cartilage through the actual perichondrium, we developed a permeable nanofiber well, which improved the efficiency of differentiation into supercartilage by realizing a homogeneous cartilage differentiation environment in stem cell spheroids. "The research results have the potential to be an effective regenerative treatment method for irreversible and intractable cartilage diseases such as degenerative arthritis," he said.

Meanwhile, the technology developed through this research is being transferred to Celloid Co., Ltd., and commercialization development is in progress through joint research with Ipcell Co., Ltd. This research was carried out with the support of the National Research Foundation's bio and medical technology development project and mid-level research project with the funding of the Ministry of Science and ICT, and the Health and Medical Technology R&D project with the funding of the Ministry of Health and Welfare.
